Xdrop®

Xdrop is a versatile and user-friendly instrument for preparing living mammalian or microbial cells, organelles, DNA, or other biological material for high-resolution downstream analyses.

Using our Xdrop Cartridges, Xdrop safely and rapidly encapsulates the living cells, organelles, or DNA fragments together with assay chemistry in highly stable, picoliter-volume, double-emulsion droplets. These droplets are stable through pipetting, vortexing, incubation, flow cytometry, sorting, and even long-term storage. What's more, cells can be recovered from the droplets for expansion.

Together with appropriate analytics technologies, Xdrop applications include:

  • Cell killing by human immune cells
  • Cytokine secretion by mammalian cells
  • Enzyme secretion by microbial cells
  • Verification of gene edits, including assessment of unintended
    on- and off-target rearrangments

With Xdrop, researchers can more easily reach high-resolution insights into the cellular functionality and genomics of humans, animals, plants, and microorganisms.

Xdrop can also generate single-emulsion droplets for other workflows, including to encapsulate DNA for unbiased whole genome amplification.

Our newest cartridge is designed to produce highly stable, ~100-picoliter, double-emulsion droplets to encapsulate mammalian cells, transforming bulk-cell into single-cell assays. Workflows using the Xdrop DE50 Cartridge include cytokine secretion and cell killing assays. Simply load the reagents and samples, put the cartridge in Xdrop, and run the program.

How is Xdrop Sort different from Xdrop?

Both instruments run targeted DNA enrichment workflows  to prepare genomic material from large libraries for screening. They can both also encapsulate living cells for a range of cell-based applications.

Xdrop Sort uniquely enables DNA or cell encapsulation and high-throughput sorting. This enables the screening of large, complex gene libraries with unprecedented speed. 

Xdrop® Sort

This new member of the Xdrop family can both generate and sort double-emulsion droplets in a user-friendly workflow that does not require any experience with fluorescence-based cell sorters.

The workflow starts with capture of 100 kb DNA fragments, small single cells, or other biological material in highly stable droplets for assays like PCR and enzyme activity assessment. Then, based on content fluorescence, droplets are sorted to obtain only the content of interest for high-resolution downstream analyses such as:

  • Verifying CRISPR edits without PCR bias that hides unintended rearrangements
  • Identifying viral and transgene integration sites
  • Assessing enzyme activity at the single-cell level
  • Performing high-throughput GFP screening

Technology

Xdrop generates two sizes of double-emulsion droplets: DE20 and DE50. The Xdrop DE20 Cartridge generates DE20 droplets, which are ~20 µm in diameter (volume: ~1.6 picoliters) and are suitable for microbial cell workflows. The Xdrop DE50 Cartridge generates DE50 droplets, which are ~50 µm in diameter (volume: ~100 picoliters) and are suitable for mammalian cell workflows. Xdrop Sort generates DE20 droplets and can also sort them based on fluorescence signals.
